The increasing variety of protein biotherapeutics being developed and the subsequent requirement for additional data for decision making early in the lifecycle is changing the landscape of analytics. Mass Spectrometry is becoming integrated into more aspects of the overall analytical control strategy for increasingly complex molecules, as well as demands for throughput to be higher and quicker with better data quality.

As a CDMO, Lonza utilises an array of MS instruments to analyse the full range of molecule types requested by our customers. We required a vendor neutral MS processing software to ensure continuity of data reporting for both internal and external customers, with the flexibility to adapt to different molecule classes. As the molecules we analyse diversify away from standard mAbs, there is an increasing requirement for data to direct decisions earlier in the product lifecycle. Together with a more MS focussed approach to IND and BLA submissions, we have seen an exponential increase in the volume of MS data being generated over the last few years. We selected Protein Metrics as the software solution to address this MS processing challenge, with the volume of data and high throughput requirements seeing us outgrow the initial installation of the Byos desktop application, and start the move to Byosphere.

Learnings:

  1. How the flexibility of data acquisition, processing and reporting is crucial to be able to adapt quickly when analyzing many different molecule types

  2. How customized base workflows which can then be tailored to customer’s specific molecules provide continuity of approach for analysis, whilst allowing product or purpose specific adaptation

  3. The data handling and processing approach that works for a low throughput of data may require re-evaluation when the load increases. A smart software solution may help to increase efficiency by eliminating the deadtime from moving large volumes of data across the network and analysis job prioritisations.
